Russian to English: Nanoparticle Size Determination Using a Near Field Scanning Optical Microscope

Предел разрешения в классической оптике определяется согласно критерию Аббе [1] длинной волны излучения и расстоянием между исследуемым объектом и объективом. Для того, чтобы преодолеть этот предел, необходимо приблизить объектив к образцу на расстояние, меньшее длины волны. До 80-х годов 20-го столетия это было невозможно.
После создания туннельного электронного микроскопа стало технически возможным приблизить и удержать объектив над образцом на расстоянии, меньшем длины волны. Таким образом, в оптической микроскопии возникло новое направление – NSOM микроскопия [2,3] (near-field scanning optical microscopy).
В данной работе исследована поверхность тонких пленок образцов раствора наночастиц активного биологического препарата ПДЕ. Для удобства исследования исходный раствор разводили в дистиллированной воде в соотношении 1:10, 1:100, 1:1000, 1:10 000, предполагая получить образцы, в которых частицы располагаются друг от друга на расстояниях, превышающих их размеры. Капля каждого раствора наносилась на предметное стекло размером 5х5 мм и высушивалась. Исходный препарат и раствор с разведением 1:10 образовывали видимую глазом пленку.
Об этом же свидетельствует и то, что время высыхания у них было заметно больше, чем у образцов с большим разведением. Пример изображений поверхности тонкой пленки образца приведен на рис.1. На изображениях образца с разведением 1:10 000 не было выявлено никаких особых структурных образований. Это объясняется тем, что частицы находятся друг от друга на расстояниях, превышающих область сканирования. Наиболее информативные оказались изображения, полученные от образцов с разведением 1:100 – частицы располагались друг от друга на расстояниях, не превышающих свои размеры. На основе полученных изображений была построена гистограмма распределения числа частиц по размерам (рис. 2). Из гистограммы видно, что в растворе активного препарата ПДЕ в основном находятся частицы размеров 90 нм (бόльшая доля частиц) и 40-50 нм (меньшая доля частиц).
Вывод. Данный метод можно использовать для анализа топографических особенностей поверхности тонких пленок и оптических свойств наночастиц с разрешением порядка λ/20.
Translation - English Nanoparticle Size Determination Using a Near Field Scanning Optical Microscope

The resolution limit in classical optics is determined according to the Abbe criterion [1] by the wavelength of the radiation and the distance between the sample under investigation and the objective lens. In order to reduce this limit, it is necessary to bring the specimen and objective closer together to a distance of less than one wavelength. Until the 1980s this was impossible.
After the creation of the tunneling electron microscope, it became technically possible to place the objective at a distance above the specimen of less than one wavelength In this way a new direction in optical microscopy arose – NSOM [2,3] (near-field scanning optical microscopy)
In the present work the surface of a fine film of a solution of nanoparticles of the biologically active substance PDE was investigated. For convenience the original solution was diluted with distilled water in ratios of 1:10,1:100, 1:1000, and 1:10,000 in an attempt to obtain specimens in which the particles were separated from each other by a distance greater than the particle size. A drop of each solution was placed on a specimen slide of 5mm x 5mm and dried out. The original preparation with a 1:10 dilution could be seen as a visible film by the naked eye.
This was supported by the fact that the drying time was significantly longer than for the more dilute preparations. An example of the images of the thin film surface is shown in fig.1 No particular structural formations were found on the 1;10,000 dilution specimen. This is explained by the fact that the particles are further from each other than the size of the scan field. The most informative images were obtained from the specimen with 1:100 dilution – the particles were separated by a distance which was no greater than their size. A particle size distribution histogram was constructed on the basis of the images obtained (fig.2). It can be seen from the histogram that there are basically particles of 90nm (the majority of particles) and 40-50nm (the minority of particles).
Conclusion. This method can be used for analysis of topographical features of the surface of a thin film and the optical characteristics of nanoparticles with a resolution of the order of λ/20.
